A Study of the Effect of Lactate on the Urea-inhibited LDH Isozyme of Human Serum

Abstract
The authors investigated in the present paper the urea induced inhibition of the LDH isozymes, lactate activation with and without urea, utilizing the pooled serum of the healthy Korean males.
Being based on the present experimental data and theoretical reason, the authors applied the present analytical procedure to the diseases of liver and heart, which showed similar elevations of the serum LDH activity in order to have differential diagnostic parameter, with the following conclusions.
1. The serum LDH activity is inhibited by urea, of which magnitudes are greater in the MLDH than in the H-LDH.
2. The lactate displays an activation of the both isozymes, which could sharply be distinguished by the different in lactate depression of the urea inhibition, a difference which is observable in the H-LDH under the presence of high concentrations of lactate, while unobservable in the M1DH under the same conditions.
3. The activities of LDH with lactate as the substrate, NAD+ the cofactor with(Vi) and without (V) fifty p moles of urea were calculated to reveal the following v/vi ratios.
(1) Normal value 1.68 (2) Acute hepatitis 6.15 (3) Hepatoma 5.47 (4) Myocardial Infarction (5) Congestive heart 0.98 failure 1.24
4. It seems significant to differentiate by the v/vi ratios the diseases of the heart and liver, the former being in the vicinity of 1. 00 and the latter being over 5.00.
